چکیده

Biofluid proteomics is a sensitive and high throughput technique that provides vast amounts of molecular data for biomarker discovery. More recently, dried blood spots (DBS) have gained traction as stable, noninvasive, relatively cheap source proteomic identification in disease injury. Snake envenomation responsible significant morbidity mortality worldwide; however, much remains unknown about the systemic response to acquiring biological samples analysis major hurdle. In this study, we utilized DBS acquired from case lethal rattlesnake determine feasibility discovering biomarkers associated with human envenomation. We identified proteins were either unique or upregulated envenomated compared non-envenomated evaluated if physiological pathways protein markers correspond observed syndromes triggered by could be detected. demonstrate provide useful information on processes resulted find evidence massive inflammatory cascade, combined coagulation dysregulation, complement system activation, hypoxia apoptosis. also detected potential indicative anaphylaxis, cardiac arrest, brain death. Ultimately, has stable pathways, which particularly relevant low-resource areas may lack materials biofluid processing storage.

Feasibility of detecting human immunodeficiency virus type 1 drug resistance in DNA extracted from whole blood or dried blood spots.

Due to high cost, availability of human immunodeficiency virus type 1 (HIV-1) drug resistance testing in resource-poor settings is still limited. We therefore evaluated the usefulness of viral DNA extracted from either whole blood or dried blood spots (DBS). Samples were collected from 50 patients receiving therapy and 10 therapy-naïve patients. Amplification and sequencing of RNA and DNA was p...

DrieD BlooD SpotS

The multi-region hybridization assay (MHAbce) for genotyping HIV-1 subtypes B, C and circulating recombinant form (CRF01_AE) was evaluated on paired plasma and dried blood spots (DBS) collected from 68 HIV-1 infected individuals in Thailand. CRF01_AE was the predominant subtype identified using plasma samples (51/62) and DBS (24/27). There was no discordance in subtype designations between plas...

Genomic Dna Extraction from Dried Blood Spots

We have developed an efficient and simple method for extracting and purifying genomic DNA from dried blood stored on filter paper. The quality of the genomic DNA extracted is tested by PCR amplification of a 255-bp fragment of the PAX8 gene sequence and the PCR products are determined for further genetic studies by single strand conformation polymorphism (SSCP) analysis. Larger DNA sequences of...

Measurement of gonadotropins in dried blood spots.

We describe direct immunofluorometric assays for luteinizing hormone (hLH) and follicle-stimulating hormone (hFSH) in fingerstick blood spots dried on filter paper, based on modifications of commercially available kits. Determinations are made from 2.5-mm-diameter discs (3 microL of dried blood) punched out from blood spot standards and samples. Sample dose detection limits of the assays (IU/L)...

N-Glycan profiling of dried blood spots.

Serum N-glycan profiles for use as clinical biomarkers of disease(s) is of increasing scientific interest. Promising profiles have already been identified in several diseases, including cancer, Alzheimer's, and diabetes. Venipuncture is routinely performed to collect the blood necessary for this type of analysis, but blood from a fingerstick placed on filter paper (known as dried blood spots (D...
